Chemists invent powerful toolkit, accelerating creation of potential new drugs

Scientists at The Scripps Research Institute (TSRI) have invented a set of chemical tools that is radically simplifying the creation of potential new drug compounds.

Pharmaceutical chemists frequently seek to generate dozens or even hundreds of variations of a given compound to see which works best. The new toolkit?described in the November 28, 2012 issue of the journal Nature?makes this process easier and cheaper, enabling previously time- and cost-prohibitive chemical modifications. This fundamental innovation is already being adopted by drug companies and is expected to speed the development of new compounds in other industries as well.

"Feedback from companies that have started to use this toolkit indicates that it solves a real problem for them by boosting their chemists' productivity and by expanding the realm of compounds that they can feasibly generate," said Phil S. Baran, PhD, a professor in the Department of Chemistry and a member of the Skaggs Institute for Chemical Biology at TSRI who led the work.

Robert Lees, PhD, of the National Institutes of Health's National Institute of General Medical Sciences, which partially funded the work, added, "Methods to predictably, selectively and efficiently functionalize a given C-H site are difficult to invent but highly significant for developing new therapeutic and diagnostic agents, and studying biomolecules. Practical chemistry for introducing fluorine atoms into organic compounds is also an important current need in medicinal chemistry. The new reagents developed by Professor Baran address both goals, and they will likely be widely utilized in both academic and industrial laboratories."

A Common and Useful Structure

The toolkit is a set of newly invented chemicals and methods for attaching functional groups of molecules to a series of common chemical structures known as nitrogen-containing heterocycles?flat molecules with rings made of carbon atoms and at least one nitrogen atom. Though their name might seem esoteric, these structures are common in both natural and synthetic chemicals, and they are found in most drugs that are taken in pill form.

"Ironically, the properties that make nitrogen-containing structures so useful in medicines are what also make them so hard to modify," said Baran. The resistance of nitrogen heterocycles to modification by traditional techniques has slowed drug discovery and has put many potential modifications out of reach.

But Baran envisioned a different scenario. "The ideal for discovery chemists would be a method that works in water, in an open flask, with procedures that are simple enough to be automated," he said.

A few years ago Baran began to glimpse the possibility of achieving such a method. His lab had just succeeded in synthesizing a complex natural nitrogen heterocycle, palau'amine, a toxin made by sea sponges in the Western Pacific that has shown anticancer, antibacterial and antifungal pharmaceutical promise. "As we developed an understanding of how that compound reacts, we recognized that it might help us solve this larger problem that discovery chemists face," he said.

The nitrogen atoms in heterocycles are what make them so tricky to handle. Traditional methods of modification typically aim to block their influence before the addition of any modifying molecules, but this approach can be cumbersome and expensive. The experience with palau'amine, which features an extremely challenging nine nitrogen atoms per molecule, led Baran and his team to try to find chemical reagents?tools, in effect?that would modify heterocycles directly.

A More Direct Route

Direct methods already had existed, but they often require extreme temperatures as well as expensive and hazardous reagents. During 2010 and 2011, Baran's lab experimented with several comparatively safe chemical reagents that can work in mild conditions to make commonly desired heterocycle modifications, such as the addition of a difluoromethyl group. One of these new reagents, a zinc dialkylsulfinate salt (DFMS) designed to transfer the difluoromethyl group, turned out to work particularly well. "We quickly realized that we might be able to make related zinc sulfinate salts that would attach other functional groups to heterocycles," Baran said.

In the new report in Nature, Baran and his colleagues describe an initial toolkit consisting of ten of these zinc-based salts, each of which attaches a different functional group to a heterocycle framework. "We selected these groups because they are commonly used by medicinal chemists," said Fionn O'Hara, PhD, a postdoctoral researcher in the Baran laboratory who was a first author of the new report with postdoctoral researcher Yuta Fujiwara, PhD, and technician Janice A. Dixon.

"In many cases, chemists will be able to use these reagents sequentially to make more than one modification to a starting compound," noted Fujiwara. The groups that can be attached with the new reagents include trifluoromethyl, difluoromethyl, trifluoroethyl, monofluoromethyl, isopropyl and triethylene glycol monomethyl ether.

To demonstrate the ability of the reagents to work in biological media, Baran's team used them to difluoromethylate or trifluoromethylate heterocycles in a solution of cell lysate?the contents of cells?as well as to serve as a buffer medium, Tris, commonly used in lab-dish tests. To underscore the general robustness of the reagents, the team even carried out such modifications in a paper cup containing Oolong tea.

Baran's laboratory collaborated in the research with scientists from the pharmaceutical company Pfizer Inc. "They provided insight into the types of compounds that would be valuable, assistance with optimization, and, most importantly, testing of the chemistry in their drug discovery laboratories, where it is meant to be used," Baran said.

The first of the zinc sulfinate salts, DFMS, now also known as Baran difluoromethylation reagent, is already being manufactured in bulk and marketed by chemical suppliers, including Sigma-Aldrich Corporation. "There has been a lot of demand for that product from the first day we listed it, which is very rare," said Troy Ryba, manager of academic chemistry strategy at the firm Sigma-Aldrich.

Baran currently is working to expand his lab's initial toolkit to provide more heterocycle-modifying choices to chemists in the pharmaceutical industry and beyond. "We have a whole slew of new reagents in the pipeline," he said.

JOHANNESBURG (AP) ? A court in South Africa has ruled that a government minister "unreasonably delayed" a decision about whether to grant the Dalai Lama a visa for a trip he planned here in 2011.

The Supreme Court of Appeal issued its judgment Thursday in the case involving former Home Minister Nkosazana Dlamini-Zuma. The court said that it was clear officials worried a visit by the Dalai Lama would upset China, which accuses the Dalai Lama of being a separatist.

The court said officials showed "deliberate procrastination" in delaying their decision.

The Dalai Lama was to attend the 80th birthday party of a fellow Nobel laureate, Archbishop Desmond Tutu. At the time, Tutu said the African National Congress-led government was worse than the country's former oppressive white regime for not issuing the visa.

Since Nov. 6, Mitt Romney has seen his Facebook followers dwindle, and his merchandise land in the discount bin. Now, a professional wrestler with a Romney/Ryan logo facial tattoo is planning to remove it, calling Romney's post-campaign performance "pretty shameful."

Eric Hartsburg told Politico the final straw came when the former GOP presidential candidate attributed his loss to President Barack Obama's "gifts" to minorities.

?It stands not only for a losing campaign, but for a sore loser,? Hartsburg said. ?He?s pretty shameful as far as I?m concerned, man. There?s no dignity in blaming somebody else for buying votes and paying off people. I can?t get behind that or stay behind that.?

Hartsburg, who said he hoped the 5-by-2-inch tattoo would make politics more fun, had initially resigned himself to keeping the mark.

?I?m the guy who has egg all over his face, but instead of egg, it?s a big Romney/Ryan tattoo. It?s there for life,? he told Politico after the election, saying he was a man of his word.

But the 30-year-old is now planning to fly to Los Angeles to have the tattoo removed, a year-long process. He hasn't ruled out getting another tattoo, maybe in 2016 when the "R" could do double duty for a potential Marco Rubio candidacy. Or, maybe, he said, he'll just get a yard sign next time.

U.S. for-profit colleges spend big on marketing while slashing other costs

(Reuters) - Google's biggest advertiser is neither a bank nor a retailer.

It's the for-profit University of Phoenix, which has recently been spending nearly $400,000 a day on ads, according to search analytics firm SpyFu, more than any financial firm or retailer, the traditional big spenders on online advertising.

That kind of spending may seem surprising coming from a college, but marketing has become vital for the university and its for-profit rivals as enrollments plummet and they fight back against a host of criticisms, including low job-placement rates.

Colleges such as University of Phoenix, the industry leader owned by Apollo Group Inc, will not only have to boost enrollments to reverse their fortunes, analysts say. They will also need to consider cutting tuition fees as well as continue to slash costs and take market share from rivals.

Operators of other for-profit colleges, whose ranks include the Washington Post Co's Kaplan business, DeVry Inc and ITT Educational Services Inc, are also boosting their spending on marketing and are among the 25 biggest advertisers on Google.

But no one is spending like the University of Phoenix, which doubled its spending on Google ads to about $380,000 per day on average between October 12 and November 12, compared with $170,000 a day in the previous month, according to SpyFu data - which Apollo describes as "gross speculation."

"I have witnessed several versions of this cycle but none as extreme as this," said Trace Urdan, an analyst with Wells Fargo Securities, who has been covering the U.S. for-profit education industry for about 15 years.

"We are going to see more pointed efforts at marketing and more price competition in an effort to try to capture more market share both from each other as well as from traditional schools," Urdan said.

Increased marketing alone will not be enough to fatten fast-shrinking profit margins and increase enrollments, however. Lower tuition fees and increased specialization of the type of programs offered, along with further streamlining of operations, will also be necessary, analysts say.

Industry bellwether University of Phoenix, which offers courses at about 230 campuses as well as online, announced plans last month to shut about half its locations and cut 800 jobs in order to save about $300 million a year by 2014.

New enrollments in the Apollo system are down nearly 50 percent in the past two years. As of August 31, enrollment totaled about 328,000.

Career Education Corp, which owns American InterContinental University and the Le Cordon Bleu colleges, and Lincoln Educational Services Corp have also announced closures.

LOW-COST MODEL

The $25 billion industry, which typically serves adults looking for a career change or a program to enhance job skills, is reeling after government investigations revealed fraud related to financial aid, worryingly high student debt loads and low rates of graduation and job placement.

"Many for-profit colleges make decisions that prioritize their bottom line, even when those decisions limit their students' opportunities for academic success," a U.S. Senate report said earlier this year.

Tuition fees, and therefore profits, is one area under pressure as potential students need to be convinced to take out loans in an uncertain job market.

Apollo, whose stock has lost about 65 percent of its value this year, implemented a tuition freeze earlier this year and promised students it will not increase prices through the course of their programs.

Apollo is also looking at different cost models, with a view to serving segments of the population that it cannot serve with current University of Phoenix tuition prices.

"We have certainly seen a lot more competition at the lower end of the price scale, and that's something we are focusing on," Apollo spokesman Mark Brennar said, while declining to offer specifics.

Wells Fargo's Urdan said it is likely that Apollo wants to compete in the low-cost end of the market by building a second brand, which it would likely do by acquiring another college rather than starting from scratch.

As colleges lower their revenue base by cutting tuition fees even as they spend more on marketing, lower margins could become the norm, analysts say. That has spooked investors already worried about sliding enrollments.

The S&P 1500 Education Services index has lost three-quarters of its value since April 2010, including a 50 percent decline in 2012.

Some for-profit colleges already differentiate themselves in the crowded higher-education market by offering programs in a particular field or by targeting students of a particular background, and that trend could accelerate.

American Public Education, for example, is known for enrolling those who work in the military and public services, while Universal Technical Institute offers programs related to the automotive industry.

For-profit colleges play up their links to employers to attract students who may otherwise opt for traditional or community colleges, said Rob Lytle, head of the education practice at advisory firm Parthenon Group.

"They are about getting people workforce employability skills, and I think they are going to be focusing tighter on that," said Lytle.

5 Things You Should Know About Gay Conversion Therapy

A therapy that claims to convert gay people into heterosexuals is being tested in court with two new cases, one of which seeks to sue counselors offering the therapy and the other seeking to defend them.

Gay conversion therapy, as it is known, supposedly helps gay people overcome same-sex attractions. But mainstream psychologists say the therapy is ineffective, unethical and often harmful, exacerbating anxiety and self-hatred among those treated for what is not a mental disorder.

Here are five things you need to know about the therapy and the current lawsuits.

1. What's happening in the courts?

Two new legal challenges are targeting conversion therapy. The first is a civil suit in New Jersey in which four former clients of a counseling group called Jonah are suing for deceptive practices. The patients argue they paid thousands of dollars for therapies that did not rid them of same-sex attractions, and that they then had to pay for mainstream therapy to repair the damage done by the conversion therapy. In a second case in California, a federal judge is hearing arguments against a new state law that bans conversion therapy for minors. The bill was signed into law in September. Conservative legal groups claim the law is a violation of the right to free speech, freedom of religion and privacy.

2. What happens in conversion therapy?

Because conversion therapy is not a mainstream psychological treatment, there are no professional standards or guidelines for how it is conducted. Early treatments in the 1960s and 70s included aversion therapy, such as shocking patients or giving them nausea-inducing drugs while showing them same-sex erotica, according to a 2004 article in the British Medical Journal.

Other methods included psychoanalysis or talk therapy, estrogen treatments to reduce libido in men, and even electroconvulsive therapy, in which an electric shock is used to induce a seizure, with side effects such as memory loss. [7 Absolutely Evil Medical Experiments]

More recently, people who have been through conversion therapy report talk therapy that emphasizes pseudoscientific theories, such as the idea that an overbearing mother and a distant father make a child gay. In an April 2012 essay in The American Prospect, writer Gabriel Arana describes his "ex-gay" therapy experience. His therapist blamed his parents for Arana's homosexuality, and urged him to distance himself from his female best friends.

Chaim Levin, one of the men suing Jonah for deceptive practices, says that he quit conversion therapy after his therapist had him strip down and touch himself to "reconnect with his masculinity," according to the New York Times. ???????????

3. Why psychologists say conversion therapy doesn't work

Homosexuality is not considered a mental disorder, so the American Psychological Association (APA) does not recommend "curing" same-sex attraction in any case. Instead, societal ignorance, prejudice and pressure to conform to heterosexual desires are the real dangers to gay people's mental health, according to a 1997 statement on "conversion" or "reparative" therapy by the APA.

A 2009 APA task force found that conversion therapies, despite being touted by religious organizations, have little evidence to back them up. A review of studies from 1960 to 2007 found only 83 on the topic, the vast majority of which did not have the experimental muscle to show whether the therapies achieved their stated goals. (Many of the people studied in the early years were court-mandated to take the therapies, adding a coercive element to those outcomes.)

The best-quality studies were more recent and qualitative, the APA task force found, meaning they focused not on the statistical effectiveness of treatment, but of the subjective experience.

"These studies show that enduring change to an individual's sexual orientation is uncommon," the task force wrote in their 2009 report. The participants continued to report same-sex attractions after the conversion therapy, and were not significantly more attracted to the opposite gender.

These studies did find that conversion therapy could be harmful, however. Negative effects included "loss of sexual feeling, depression, suicidality and anxiety."

4. How did conversion therapy get started?

The desire to turn gay people straight goes way back. In 1920, Sigmund Freud wrote of a lesbian patient whose father wanted to see her converted to heterosexuality. Freud echoed modern psychologists by responding that changing sexual orientation was difficult and unlikely. He offered to see the woman anyway, but later broke off the therapy due to her hostility. In 1935, Freud went even further, writing to a woman who wanted her homosexual son converted that homosexuality "is nothing to be ashamed of, no vice, no degradation; it cannot be classified as an illness."

Other psychologists throughout the early mid-1900s believed homosexuality could be changed and recommended a variety of treatments. One of the stranger attempts was an effort by Viennese endocrinologist Eugen Steinach to transplant testicles from straight men into the scrotums of gay men in an attempt to rid them of same-sex desires. It didn?t work.

One of the most prominent advocates of conversion therapy in the 1940s and 50s was Edmund Bergler, who saw homosexuality as a perversion and believed he could "cure" gay people with a punishment-based, confrontational therapy style.

Once the American Psychiatric Association stopped classifying homosexuality as a mental disorder in 1973, conversion therapies lost support. But religious-right organizations such as Exodus International and Focus on the Family's Love Won Out took up the charge, promoting their own "ex-gay" therapies. A small group of psychologists, splitting with their peers, continue to promote the therapies, founding the conversion therapy organization NARTH, or the National Association for Research & Therapy of Homosexuality. The group has religious links; for example, one of its founders and former president, psychologist Joseph Nicolosi, is a one-time spokesman for Focus on the Family. 5. Okay, but what about that one study that found conversion therapies work?

Groups that promote conversion therapy often point to a single study to support their work. In 2003, famed psychiatrist Robert Spitzer, who spearheaded the removal of homosexuality from the American Psychiatric Association's mental disorder list in 1973, reported in the journal Archives of Sexual Behavior that interviews with conversion therapy patients suggested that some people could change their sexual orientation.

The paper was incendiary and highly criticized, given that it relied on interviews with patients instead of measurable benchmarks of same-sex desires. Conservative groups were delighted to have support from Spitzer, who wasn't tainted with religious bias or anti-gay ideology; gay organizations felt betrayed.

In the end, however, Spitzer came to agree with his critics. There was no way to confirm that what his interviewees said was true, he wrote in 2012 to the editor of the journal Archives of Sexual Behavior. The study, he said, was fatally flawed.

"I believe I owe the gay community an apology for my study making unproven claims of the efficacy of reparative therapy," Spitzer wrote.

    New mechanism for cancer progression discovered

    ScienceDaily (Nov. 27, 2012) ? The protein Ras plays an important role in cellular growth control. Researchers have focused on the protein because mutations in its gene are found in more than 30 percent of all cancers, making it the most prevalent human oncogene.

    University of North Carolina Lineberger Comprehensive Cancer Center and Harvard researchers have discovered an alternative mechanism for activating Ras that does not require mutation or hormonal stimulus. In healthy cells, Ras transmits hormone signals into the cell that prompt responses such as cell growth and the development of organs and tissues. A mutation on the RAS gene can chronically activate those signals, leading to tumor initiation and progression.

    In an article published on-line in a November issue of Nature Structural and Molecular Biology, the UNC and Harvard teams discovered that modification of Ras at a specific site with a small protein known as ubiquitin can also lock Ras into an active signaling state. Thus, modification of Ras with a single ubiquitin -- a process known as monoubiquitination -- switches Ras to an active signaling state by disrupting the action of another protein known as the GTPase activating protein, or GAP. Work by two of the papers co-authors, Atsuo Sasaki and Lewis Cantley of Harvard, had previously found evidence for Ras's potential to become activated and promote Ras-mediated tumorigenesis by monoubiquitination.

    Because of the strong link between Ras and cancer, Ras should be an attractive target for drug discovery efforts. Despite considerable efforts at developing treatments targeting the protein, Ras itself is now considered to be 'undruggable', leading researchers to try new approaches to developing drugs that target activated Ras. This could lead to benefits beyond cancer therapies, as the RAS gene has also been linked to developmental disorders such as Noonan syndrome, Costello syndrome and autoimmune lymphoproliferative syndrome.

    Lead researcher Rachael Baker, a PhD candidate doing joint work in the labs of Henrik Dohlman, PhD, professor of pharmacology and vice chair of biochemistry and biophysics and Sharon Campbell, PhD, professor of biochemistry and biophysics at UNC, developed a novel method to modify Ras with ubiquitin and then subsequently characterized how ubiquitin modification can lead to Ras activation.

    The attachment of ubiquitin to Ras at a specific site leads to Ras activation, much like with an oncogenic mutation, leading to an increased potential for cancer formation. Baker notes that the reaction can be reversed by enzymes in the cell that remove ubiquitin, making these enzymes possible targets for future pharmaceutical research.

    "Establishing how Ras is activated by ubiquitin is just the first step in understanding this novel mechanism of cellular regulation." said Campbell.

    The researchers next step will be to obtain a more detailed understanding of its role in cancer progression, first in cells and in animals and eventually in cancer patients.

    Exec embroiled in HP-Autonomy mess lashes out

    (AP) ? An executive vilified by Hewlett-Packard Co. on allegations of an accounting ruse is escalating his counter-attack on the company accusing him.

    In a letter to HP's board released Tuesday, former Autonomy CEO Mike Lynch fiercely defended his integrity. He also questions whether HP is using the charges of financial shenanigans to cover up damage caused by its own ineptitude.

    HP fired back with a statement that predicted its findings of misconduct will be proven in anticipated legal proceedings about the dispute.

    Lynch's missive represented his most extensive remarks since HP accused him last week of deceiving the company about the growth of Autonomy, a business software maker that HP bought for $10 billion last year. He had previously denied HP's accusations in interviews with various news organizations.

    HP absorbed an $8.8 billion charge to reflect Autonomy isn't worth what the company paid. About $5 billion of that charge stemmed from improper accounting, according to HP.

    "Can HP really state that no part of the $5 billion write down was, or should be, attributed to HP's operational and financial mismanagement of Autonomy since the acquisition?" Lynch wondered in his letter. He demanded that HP's board provide more specifics about how it arrived at its calculations.

    The Autonomy mess has deepened a steep decline in HP's stock price, which has cut the Palo Alto, Calif., company's market value in half since the beginning of the year. HP had already been struggling because its personal computer and printer businesses have been faltering as more people buy smartphones and tablet computers. HP's stock fell 38 cents Tuesday to close at $12.36.

    HP CEO Meg Whitman said last week that she fired Lynch in May because Autonomy wasn't hitting its financial targets. After Lynch departed, Whitman said a whistleblower triggered an extensive investigation that culminated in HP's conclusion that it had been duped before it agreed to buy the British company. HP said it has handed its findings over to securities regulators in the U.S. and the U.K., as well as the U.S Justice Department.

    Even if authorities don't file charges, HP said it plans to take legal action against the people it believes manipulated Autonomy's finances.

    "We look forward to hearing Dr. Lynch and other former Autonomy employees answer questions under penalty of perjury," HP said in a statement. The honorific in HP's statement refers to Lynch's doctorate in computing.

    HP, meanwhile, faces at least one shareholder lawsuit. On Monday, a stockholder who owns 200 shares accused HP of concealing problems with Autonomy and other acquisition.

    Lynch said he wished he had been given a chance to help "make HP great again. I have been truly saddened by the events of the past months, and am shocked and appalled by the events of the past week."

    (AP) ? They share a city, first place and now the belief that this new-look, New York matchup could be a real rivalry.

    And what a stirring start to it for the Nets and Knicks.

    The Nets won the crowd ? and the game ? in the first meeting in Brooklyn, pulling out a 96-89 overtime victory on Monday night to tie the Knicks atop the Atlantic Division on a breakthrough night for their franchise.

    "It was absolutely fantastic. It was a great game for the fans," Nets center Brook Lopez said. "Having two great teams in New York, the fans are in the best position of all."

    With their fans outnumbering and at times outchanting the Knicks' counterparts for a change, the Nets improved to 7-1 in their new home before a sellout crowd of 17,732 that was more black than blue, just the way the Nets hoped.

    "Every time some sort of Knicks contingency started to cheer, our fans got louder," Nets coach Avery Johnson said.

    Jerry Stackhouse, wearing the No. 42 Jackie Robinson made famous in Brooklyn, hit the tiebreaking 3-pointer with 3:31 left in overtime as the Nets controlled the extra period of a playoff-like game that neither team led by more than seven.

    Lopez had 22 points and 11 rebounds, and Deron Williams added 16 points and 14 assists as the Nets joined the Knicks at 9-4 amid a loud chant of "Brook-lyn! Brook-lyn!" at the finish. Stackhouse scored 14 points.

    Carmelo Anthony had 35 points and 13 rebounds, but was only 10 of 16 at the free throw line and missed the potential go-ahead jumper with about 5 seconds left in regulation. Tyson Chandler finished with 28 points and 10 boards, but no other Knicks player was in double figures.

    "They're across the bridge. They're here in Brooklyn. We're in the city. They're in the same division, the same sports town. I guess you can say this started something," Anthony said.

    Raymond Felton was just 3 of 19 from the field, but the Knicks had to stick with him with fellow point guard and former Nets star Jason Kidd out with back spasms. Another starter, Ronnie Brewer, was lost for the overtime with a dislocated left ring finger.

    "I'm upset at myself. I had a bad shooting night," Felton said. "I'm probably just as mad as any fan, any player, anybody you could think about."

    The first matchup of Brooklyn and New York pro franchises since the Dodgers played the New York Giants on Sept. 8 1957, at the Polo Grounds was loud, energetic, and close throughout. Both teams played as if this meant more than the usual regular-season game.

    "It was just a total 180 from what we saw last year, where it was mostly Knicks fans and all the chants and all the cheers were for them. It's great to feel that we have a home-court advantage finally," Williams said.

    "This was a playoff-type atmosphere. You could definitely feel it on the court and it was a great game to be a part of."

    It's the latest the Nets have had at least a share of first place since Jan. 30, 2007, when they and Toronto were both 22-23. It's the latest the Knicks and Nets have been tied atop the division since Nov. 30, 1997.

    The game was originally scheduled as the Nov. 1 season opener for both teams but was postponed after Superstorm Sandy, when much of the mass transit around the city was knocked out. Barclays Center sits above a major transportation hub, and without subways running many fans wouldn't have been able to get here.

    The Knicks have always been the clear No. 1 franchise in the area, even when the Nets had Kidd and were better, and the Nets have long believed the move to Brooklyn and the $1 billion Barclays Center would help them even things. Nationally televised and with more than 100 media members, this was easily their most anticipated game in years, though Johnson denied that it meant more to the Nets than the Knicks.

    "I think it's just as important for them, because if we're so much the new kid on the block and we're trying to come in and gain some territorial rights, I think they're going to try to do everything they can to push back, so I think it's great," Johnson said before the game. "I don't think it's so much of a Duke-North Carolina rivalry yet, but hopefully one day it'll get there."

    Johnson estimated that games in New Jersey since he'd been with the Nets were an 80-20 fan split for the Knicks, and he hoped that would turn around Monday. It didn't flip completely, but it seemed the home team's fans made up about 60-65 percent of the crowd, the orange and blue that was long so ubiquitous harder to spot this time.

    But Johnson made it clear the Nets needed to win the game, not just the crowd, for this to be a successful night, and his team was in great position up five with 4 minutes to play in regulation. The Knicks ran off eight straight to take an 84-81 lead with 1:40 left, but they wouldn't score again. Lopez tied it with a free throw with 24 seconds remaining, and after Anthony missed a jumper, Williams was off on a long attempt before the buzzer.

    Chandler dunked to open overtime, but the Nets answered with eight straight points, taking the lead for good at 89-86 on Stackhouse's 3-pointer with 3:31 remaining before three straight points from Gerald Wallace, who finished with 16.

    Both sides downplayed the idea of a rivalry ? yet ? leading into the game, but change was noticeable right from the start.

    The Knicks were loudly booed during the announcement of starting lineups, with Anthony, a Brooklyn native whom the Nets tried to acquire before the Knicks did, receiving the type of loud, mixed response that usually goes to LeBron James.

    There were dueling chants on one early possession, Knicks fans hollering "Defense! Defense!" while Nets supporters tried to drown them out with "Brook-lyn! Brook-lyn!"

    The only time crowd support seemed unanimous was when local sports stars or celebrities, such as Jay-Z and Beyonce sitting near the Nets bench, were shown on the overhead scoreboard.

    Notes: The Nets have won six straight at home for the first time since December 2003. They started 18-1 at home in 2002-03. ... The teams meet three times in the next 3? weeks. The Knicks return to Brooklyn on Dec. 11 and host the Nets on Dec. 19. ... With Kidd out, the Knicks started Kurt Thomas ? and actually got older. Kidd will turn 40 in March. Thomas, the league's oldest player, turned 40 in October. ... Anthony (10.9 ppg) and Lopez (7.9) entered as the NBA's top two first-quarter scorers. Anthony had only four points, while Lopez scored eight.

    Infected and unaware: HIV hitting America's youth

    CHICAGO (Reuters) - More than half of young people in the United States who are infected with HIV are not aware of it, according to a report by government health officials that zeroes in on one of the remaining hot spots of HIV infection in America.

    Young people ages 13 to 24 account for 26 percent of all new HIV infections, according to the report by the U.S. Centers for Disease Control and Prevention, which was released on Tuesday in advance of World AIDS Day on December 1.

    "Given everything we know about HIV and how to prevent it in 30 years of fighting the disease, it's just unacceptable that young people are becoming infected at such high rates," CDC Director Dr. Thomas Frieden said on a conference call with reporters.

    Every month, 1,000 young people in America become infected with HIV, an incurable infection that costs $400,000 to treat over a lifetime, Frieden said. If left untreated, HIV infection leads to AIDS and early death.

    "The data are stark and worrying," Dr. Kevin Fenton, director of the National Center for HIV/AIDS, Viral Hepatitis, STD and TB Prevention at the CDC, said in a telephone interview.

    In 2010, 72 percent of the estimated 12,000 new HIV infections in young people occurred in young men who have sex with men, and nearly half of new infections were among young, black males.

    "We are particularly concerned about what is happening with HIV among young black gay and bisexual men," Fenton said. "They account for 39 percent of all new infections among youth and more than half of new infections among young men who have sex with men."

    Fenton said the proportion of young people infected with HIV has remained relatively stable during the last few years, but infection rates appear to be increasing in these populations.

    And because many of the newly infected gay or bisexual males are just beginning to explore their sexuality, stigma and homophobia are making HIV testing and treatment far more challenging.

    Although the CDC and the American Academy of Pediatrics recommend routine screening for HIV starting in the early teens, "too few young people are getting tested," Frieden said.

    Only 13 percent of all high school students and only 22 percent of sexually active high school students have ever been tested for HIV, the CDC found.

    The figures for young adults aged 18 to 24 are not much better, with only 35 percent of people in this age group reporting ever having been tested for HIV.

    Frieden said one reason for the higher rates of infection among young gay and bisexual men of all races was a higher rate of risky behaviors compared with their heterosexual peers.

    According to the report, a large analysis of risk behaviors among high school students revealed that gay and bisexual males are much more likely to have multiple sex partners, to inject illegal drugs and to use alcohol or drugs before sex. They are much less likely to use condoms.

    And because so few get tested, HIV-infected people under age 25 are significantly less likely than those who are older to get and stay in care, and to have their virus controlled at a level that helps them stay healthy and reduce their risk of transmitting HIV to partners.

    The CDC also found that many young men who have sex with men are less likely than others to have been taught about HIV or AIDS in school.

    "We have to correct a lot of myths and misconceptions," Frieden said. "It is astonishing the level of ignorance about basic physiology that may high school and middle school students have."

    To address some of the issues, the CDC is funding a program that targets both at-risk youths and the homophobia and stigma in the community that drives them underground.

    In September, Georgia, a state where new HIV infections among those 13 to 24 years old exceed the national average - accounting for as many as one-third of all new HIV infections - won a grant as part of a pilot project to find better ways of targeting these at-risk youth.

    "We think that it's really critical that the discussions we have about HIV prevention and access to HIV become fully integrated into the social fabric of the youth culture," Dr. Melanie Thompson, of the Georgia Department of Public Health, said in a telephone interview.

    Care and Prevention in the United States, known as CAPUS, is a three-year program led by the CDC and other government agencies aimed at reducing HIV and AIDS among racial and ethnic minorities in the United States. It focuses on addressing social, economic, clinical and structural factors influencing HIV health outcomes.

    "It is a huge challenge," said Thompson, "but I think if we do this from the point of view of trying to end an epidemic that is decimating our young people, and do it in a way that is science-based, I think we can make progress."

You see, before changing its name from Macrovision, Rovi acquired Gemstar-TV guide in 2008 -- which was probably most well known for the integrated program guide common in higher end TVs throughout the last decade. While most people just use the guide offered on their provider's set-top, this guide is for those who actually use the clear-QAM or over-the-air tuner in their TV. The way it worked is that manufacturers baked the software into the TV, which would then tune into a local affiliate's broadcast of a week's worth of guide data. The setup successfully made it through the digital transition -- minus the truckloads of analog-only TVs ceasing to work along the way -- as new TVs featured a digital version of the service, as well as some digital converters, like the DTVpal DVR.

    Facebook stock climbs after upgrade

    NEW YORK (AP) ? Facebook's stock is climbing after an upgrade from an analyst who believes Wall Street is underestimating the company's potential to grow revenue.

    Bernstein Research analyst Carlos Kirjner said Monday that Facebook Inc. is likely to beat Wall Street's revenue expectations over the next 12 to 24 months. He expects Facebook to hit $6.98 billion in revenue in 2013, some 9 percent higher than the average analyst's expectation. In 2011, the social network reported revenue of $3.71 billion.

    Kirjner upgraded Facebook's stock to "Outperform" from "Market Perform," saying that mobile advertising will drive revenue growth. He wrote in a note to investors that Facebook will likely be able to increase the number of advertisements its users view each day, with little chance they will see the additional ads as a "material deterioration" of their Facebook experience.

    That said, Kirjner still thinks Facebook is a risky investment, because it depends on the long-term success of social advertising ? a still new and unproven advertising model.

    Facebook's stock is up $1.85, or 7.7 percent, at $25.85. It's still down 32 percent from its initial public offering price of $38.

    (AP) ? Atlanta coach Mike Smith spoke highly of the Falcons' defense. Without its solid performance, Matt Ryan may not have had an opportunity to lead his latest game-winning drive.

    The Falcons (10-1) held off Tampa Bay 24-23 Sunday, overcoming two turnovers by their star quarterback in part because the Buccaneers settled for a pair of field goals rather than touchdowns after Ryan threw a second-quarter interception and lost a fumble on a fourth-quarter sack.

    "I thought our defense did an outstanding job again. They were put in some tough situations after turnovers," Smith said, adding that limiting Bucs rookie Doug Martin to 50 yards rushing on 21 carries was a key to the game as well.

    "We knew it was going to be one of our 'musts,' and I thought we did a nice job," Smith added. "I thought we played the run extremely well, controlled the line of scrimmage. That is an outstanding running back."

    Ryan completed his first 10 passes and finished 26 of 32 for 353 yards and one touchdown, an 80-yarder to Julio Jones, who had nine receptions for 147 yards despite playing on a sore right ankle that limited him in practice.

    Atlanta got a lift from the return of linebacker Sean Weatherspoon, who was back after missing three games with an ankle injury.

    Josh Freeman threw for 256 yards for Tampa Bay, however he was sacked twice and failed to throw a touchdown pass for the first time in 14 games. Martin scored both of his team's TDs on 1-yard runs and Connor Barth kicked three field goals.

    "It's hard to win games by threes," coach Greg Schiano said. "Sevens add up considerably quicker."

    The Falcons (10-1) kept pace with the Houston Texans for the NFL's best record and ended a four-game winning streak by the up-and-coming Bucs (6-5).

    "They are playing with a lot of confidence," said Ryan, who has won seven of his past eight starts against the Bucs. "It's always been a tough place for us to come play. They play with great effort, and I think they have a good football team."

    Ryan teamed with Jones on his long TD midway through the third quarter, but the Falcons found themselves trailing 23-17 after the Bucs marched to Martin's second TD, then used a sack-fumble to set up a field goal that left Atlanta needing another fourth-quarter comeback to win.

    Ryan obliged by a leading a six-play, 64-yard march that Michael Turner finished with a 3-yard TD run that put the NFC South leaders ahead for good with 7:55 remaining.

    Barth missed a 56-yard goal for Tampa Bay in the closing minutes, and the winning streak that's helped the Bucs climb back into playoff contention following a 1-3 start ended when Freeman's desperation pass fell incomplete in the Atlanta end zone with no time left on the clock.

    "The last drive, that's something we've done before this season," Falcons tight end Tony Gonzalez said. "I really think that's a positive for us to be able to come up with plays like that when we have to like that. ... I'm really happy with the way we responded."

    It's getting to be a way of life. Ryan has also led late marches to beat Carolina, Washington, Oakland and Arizona this season and has put together 21 game-winning drives in the fourth quarter or overtime since entering the league in 2008 ? four of them against the division rival Bucs.

    The Falcons won't have long to celebrate this one. They host the New Orleans Saints on Thursday night in a rematch of the only game they've lost.

    "Ten and 1 is great, but I'll tell you what I've said since the beginning of the season: We're just jockeying for position," Gonzalez said. "We just want to put ourselves in the best position, playing the best football. Right around this time is when you want to start jelling as a team and take it on into the playoffs because that's the most important thing."

    The win stopped the Bucs, who've won five or seven following a 1-3 start, from tightening the NFC South standings. Turner also scored a fourth-quarter TD that helped the Falcons overcome six turnovers to beat Arizona 23-19 last week.

    "It was a hard-fought game by both teams, a very physical game," Schiano said. "But I do think missed opportunities were a big part of it. There were some chances both offensively and defensively, and in the kicking game."

    NOTES: Martin scored on a pair of 1-yard run for the Bucs. ... After throwing seven interceptions in Atlanta's first nine games, Ryan has thrown six the past two weeks. ... Falcons CBs Asante Samuel (shoulder) and Dunta Robinson (head) left the game with injuries in the third quarter, but returned.
